紫杉醇负载 EDC 交联丝素纳米粒: 结肠癌治疗的潜在方法。

Abstract:

: 结肠癌是最危及生命的癌症之一，发病率和死亡率都很高。目前的一线治疗无效，有许多不必要的效果。紫杉醇封装在纳米颗粒中的非标签使用证明了一种创新方法。在这项研究中，我们报道了用于抗癌目的的新型紫杉醇负载 EDC 交联丝素纳米颗粒 (PTX-FNPs)。采用去溶剂化法制备颗粒，并对其理化性质进行了良好的控制，包括粒径 (300-500 nm) 、 zeta 电位 (- 15 至 + 30 mV) 、药物包封率 (75-100%)，结晶度，药物溶解度 (1-10 倍增加)，溶出曲线，稳定性(在静脉稀释液中> 24 h，在 4 ℃ 下储存> 6 个月)。在体外研究中，所有制剂对红细胞没有毒性，而保留了紫杉醇对 MCF-7 乳腺癌和 Caco-2 结肠癌细胞的细胞毒性。有趣的是，PTX-FNPs 可以被 Caco-2 细胞快速摄取，因此与游离药物相比，紫杉醇的效力增加了 10 倍。图形摘要。
