SIRT6 与 SIRT5 协同通过 ampk α 信号通路调节牛前脂肪细胞分化和脂质代谢。

Abstract:

: 前脂肪细胞分化和脂质合成是肌内脂肪 (IMF) 沉积和脂质代谢稳态的关键步骤。牛肉的 IMF 含量不仅决定了肌肉和脂肪的比例，还决定了牛肉的品质、风味和感官特性。维持脂质代谢稳态是防治糖尿病、肥胖等代谢性疾病的关键手段。SIRT6 是一种 ADP-核糖基转移酶和乙酰和长链脂肪酰基的 NAD + 依赖性去乙酰化酶，在脂质和葡萄糖代谢中起核心作用, 与营养过剩和衰老引起的糖尿病和肥胖的发生密切相关。本研究基于牛前脂肪细胞分化和肥胖小鼠模型，综合利用转录组测序 (RNA-seq) 和形态学鉴定方法探讨抑制 SIRT6 对分化和脂质合成的影响，以及相关的分子机制。此外，分析了 SIRT5 和 SIRT6 对分化和脂质沉积的反馈协同调节作用。结果表明，在牛前脂肪细胞的分化过程中，抑制 SIRT5 显著促进了 SIRT6 的表达。此外，SIRT6 抑制牛前脂肪细胞分化和脂质合成，与 SIRT5 协同减少脂质沉积，抑制前脂肪细胞的细胞周期阻滞。而且，体内验证实验也获得了一致的结果。此外，SIRT6 通过激活单磷酸腺苷活化蛋白激酶 α (ampk α) 通路抑制前脂肪细胞分化和脂质沉积。上述结果为理解 SIRT6 在调节牛脂肪细胞分化和脂质代谢中的功能提供了新的方法，也为临床治疗糖尿病和肥胖提供了新的靶点。
